A single use reactor specifically designed to be a fermentor meets the performance requirements of dense, rapidly growing microbial cultures while offering the benefits of quick process setup, reduced contamination risk, and proven quality of the original single-use bioreactor. These single-use fermentors (SUF) enable production facilities to move their existing processes from CIP/SIP stainless steel fermentors into single-use without modifying their existing procedures. The manufacture lead time of the SUF hardware is a fraction of traditional vessels. As each SUF bioprocess container ships sterile and validated, down time between cultures is minimal, reducing the overall screening time and allowing for more production volume in less space. SUF having a working volume range from 6L to 300L allows for screening and optimization at 6L and then scale-up as expression systems are moved to production. Here we compare protein production in the SUF to traditional stainless steel fermentors using strains of E. coli, P. pastoris, S. cerevisiae, Bacillus subtilis, and T. album with temperature shifts, 2vvm gas flow, and seeding at 20% working volume.
