Glycoside Hydrolase (GH) family 61 protein can significantly enhance enzymatic biomass hydrolysis. This GH61 effect depends on a non metal or proteinaceous activator, such as ascorbic acid, gallic acid, or glutathione. To better understand GH61 reaction, a wide range of biomass-derivable and known enzyme cofactors were tested as potential GH61 activator.  Based on selected enzymatic lignocellulose hydrolysis assay, some key sub-structural features were noted for their ability to enable host molecules in activating GH61.
