The evaluation of the kinetics of enzymatic hydrolysis using sugarcane bagasse pretreated with alkaline hydrogen peroxide (AHP) as substrate were performed with low and high levels of enzyme loadings (1.67 – 30 FPU/g dry pretreated biomass and 7.33 – 50 CBU/g dry pretreated biomass). A model taking into account cellobiose and glucose inhibition, adsorption–desorption equilibrium of cellulase on substrate and irreversible adsorption of β-glucosidase to lignin was used. The simultaneous estimation of kinetic parameters of the model was performed using the Pikaia genetic algorithm (GA). Plackett-Burman (PB) screening design is a reliable technique, which can identify among large numbers of factors the ones that are significant for the desired response. In this work, as some parameters can have different impacts at the beginning, middle or at the end of the hydrolysis, a dynamic PB design evaluation was proposed, where the influence of the kinetic parameters on cellulose, cellobiose and glucose concentration was determined as a function of the hydrolysis time.
