Various microbial strains producing cellulases and xylanase were isolated from the rotten woods and their surrounding soils. One fungal strain producing high amount of cellulases and xylanase was isolated and identified as Lecythophora sp. NP3-6-3 according to its ITS, 18S, and 26S sequences. The enzyme production by the isolate from various lignocellulosic substrates such as flame grass, reed, empty fruit bunch, rice straw, and barley straw were examined. Among them, barley straw produced the highest level of endocellulase, exocellulase, filter paperase, and, xylanase. The optimum culture conditions were culture temperature, 36°C; initial medium pH 5; C/N ratio, 7; and culture time, 6 days; The production of CMCase, FPA, β-glucosidase, avicelase, xylanase production after the optimization was improved to 12.3 U/mL, 6.7 U/mL, 4.6 U/mL, 2.1 U/mL, 65.0 U/mL, respectively, from 4 U/mL, 2.3 U/mL, 2.3 U/mL, 1.8 U/mL, 24.1 U/mL, respectively, before optimization.
