Study on Metabolic Regulation of Spinosad Producing Strains

Spinosad is a novel kind of 12-member macrolides antibiotics produced by Saccharopolyspora spinosa, which is a safe and effective biopesticde with an excellent environmental and mammalian toxicological profile and has wide application prospect in many fields such as agriculture and stockbreeding. In this study, S. spinosa strain HB-B was matagenized in turn by acridine orange, UV irradiation, nitrogen ion implantation etc. Two mutants, a high producing strain HB-A and a low producing strain HB-C, were obtained and had hereditary stability. Understanding the regulation of spinosad biosynthesis is a prerequisite for guiding the rational reconstruction. So far, there is little report on the related regulation factors. Thus, different yield of strains HB-A, HB-B and HB-C were used as the experiment materials. The biomss were harvested at different time of the metabolic cycles, then the RNA was extracted and transcriptomic analysis was performed, intending to study the regulatory factors of differential genes. This work may provide theoretical basis for guiding metabolic regulation and will allow more strategies to improve the yield of spinosad.
