A novel immuno-stimulating agent(AABPM-0065, bio-processed glucomannan) was obtained through stepwise degradation of the cell wall components of Auricularia auricular judae fruit body by several enzymes such as mannase, glucanase, cellulase, hemi-cellulase and pectinase. Through this bioconversion process, the inactive polysaccharide components contained in the cell wall of Auricularia auricular judae was found to be converted into the valuable active agent (AABPM-0065) having the characteristics of strong immuno-stimulating efficacies. It was revealed that the major ingredients showing the effects of the non-specific immuno-stimulants are water-soluble polysaccharides, and appeared to have the structure of acetylated glucomannan. Notably, the medicinal polysaccharides were observed to help to enhance not only the innate immunity by activating macrophages, but also the acquired immunity through activation of dendritic cells. In this paper, various impressive validation results will be presented, proving the immune-activating effects of the bioconverted products through in-vitro and in-vivo tests with various immune cells and experimental mouse, respectively.
