S140: Metabolizing enzyme microarray to generate metabolites for early toxicity screening

A 3-dimensional cell culture microarray platform to screen for both acute toxicity and metabolism-induced toxicity of chemical compounds will be described. The DataChip (the Data Analysis Toxicology Assay Chip) is a miniaturized 3D cell-culture array containing 1,080 individual cell cultures in a spatially addressable format on a glass chip the size of a microscope slide. The MetaChip (Metabolizing Enzyme Toxicology Assay Chip) is a similarly configured array of immobilized metabolic enzymes designed to emulate effects of Phase I and II metabolism. The MetaChip is flexible and can accommodate liver microsomes, s9 fractions, and many combinations of recombinant metabolic enzymes. Results from combining the DataChip and MetaChip for toxicity screening will be described.
