The Use of Computational Fluid Dynamics to Enhance Monoclonal Antibody Productivity
Stefanie Brady, BioPharmaceutical Development, Human Genome Sciences, Inc., 14200 Shady Grove Road, Rockville, MD 20850
During start-up of HGS’s Large Scale Mammalian facility, it was discovered that performance of the cell culture was not optimal at the 20,000 L scale. Growth rates and product titers, although acceptable, were lower than those observed at the pilot scale. To determine the root cause and identify potential solutions, Computational Fluid Dynamics (CFD) was utilized. Scale-down pilot runs were also used to aid in verifying the cause of the problem and to validate the CFD modeling approach. This presentation will share the findings from the CFD analysis and how the analysis aided in quickly implementing solutions and effectively scaling the process to the 20,000L bioreactor.
