Isolation and Identification of Bacterium Producing Antimicrobial Substance from frog skin ( Hyla savignyr) and Extraction of its Antimicrobial Substance
Due to increasing of microorganisms resistance to antibiotics, finding new antibiotics is necessary. The aim of this study is isolation of a bacterium producing antimicrobial substance from frog skin (Hyla savignyr), aiming toward finding a new antibiotic and then its extraction, purification and identification
Materials and methods:
An antimicrobial substance producing bacterium was isolated using nutrient agar medium followed by growing in broth medium (nutrient broth and synthetic), supernatant was concentrated on the disk and finally tested on several standard microbial strains including Escherichia coli, Pseudomonas aeroginosa, Bacillus subtilis, Candida albicans, through diffusion disk. Identification of the bacterium using both Phylogenitic analysis and classic tests were done. In order to purify the polar antimicrobial substance for identification, yielded supernatant was washed with chloroform solution and ethyl acetate.
Results:
The isolated bacterium was rod-shaped, gram-positive with slime milky colonies. Phylogenetic analysis showed relationship between this bacterium and one of Bacillus subtilis strains. Antimicrobial substance derived from this bacterium has an effect on Pseudomonas aeroginosa, Escherichia coli, Bacillus subtili, Candida albicans.
Discussion:
For the first time a bacterium has been extracted from the skin of frog. Its antimicrobial effect on Bacillus subtilis and Candida albicans is more than to the Pseudomonas aeroginosa and Escherichia coli.
