Small-scale fermentation as a troubleshooting, process-insight and support tool for commercial BIO-PDO™ fermentation operations
C.J. Guske, Tate & Lyle, 2200 E. Eldorado Street, Decatur, IL 62525 and E.S. Miller Jr., DuPont Tate & Lyle Bio Products, Loudon, TN 37774.
Small-scale fermentation (≤20L in volume) is most often relegated to the realm of bench-scale bioprocess screening and development, one of the early phases of the scale up and commercialization of new products. Conventional wisdom suggests that these small vessels have little else to offer once the process moves out of the laboratory and into the pilot plant or fermentation production facility. Practical experience and ingenious strategies, however, have demonstrated otherwise, proving repeatedly the value of small-scale fermentors as a key, on-site support resource for successful startup and ongoing support of fermentation production facilities. Potential applications include validation, troubleshooting and optimization of production cultures, media and operating conditions. A simple approach and application of small-scale fermentation capabilities in a fermentation production facility will be illustrated using examples from DuPont Tate & Lyle Bioproducts LLC’s Bio-PDO™ experience.
