Creating enzymes suitable for industrial application normally requires significant improvements in stability and solvent tolerance, as well as relief of inhibition and acceptance of high substrate concentration. In our experience no one tool can be applied to address optimally all these problems. We will describe the use of shuffling, a bioinformatics program called ProSAR, and biocatalyst panels to generate ketoreductases for the production of key pharmaceutical intermediates.
