On-line monitoring of cell lysis in homogenization processes

In fermentations where the product accumulates intracellularly a cell lysis (homogenization) method is needed. It is often difficult to follow the outcome of a cell homogenization process because of the very crude nature of the homogenate that contains a high number of different cellular compounds and fines in different sizes. Additionally homogenization processes tend to be dependent on the equipment, and data obtained from laboratory scale equipment may or may not be scalable to production scale. An on-line monitoring method to observe cell lysis in homogenization processes is desirable in order to assist process development and scale-up. The parameters pH, conductivity and capacitance (viable cells) were measured before and after homogenization. The correlation of these parameters to homogenization pressure and to the release of product will be presented.
