Cubist Pharmaceuticals is addressing the considerable need for new antibiotics by developing innovative approaches to discover novel secondary metabolites from actinomycetes and fungi. This discovery engine consists of a method of high-throughput micro-titer fermentation, followed by rapid evaluation of these small scale cultures for the production of novel, potent antibacterial natural products. This poster will outline the micro-fermentation steps and bioassay guided fractionation used to rapidly screen hundreds of thousands of soil isolates collected from all over the world, and highlight some of the more interesting antimicrobial natural product compounds discovered by this method at Cubist. Promising cultures are prioritized based on activity and potency for scale-up fermentation and isolation of bioactive molecules.
