Large screening of Exopolysaccharides(EPS) Overproducers through Protoplast Formation and Bioreactor Studies on the EPS Production by Suspended Cultures of Inonotus obliquus Mycelia

Studies on the production of exopolysaccharide(EPS) from the suspended myelial cultures of Inonotus obliquus belonging to basidiomycetes fungus have been carried out. EPS biosynthesized by I. obliquus has a promising potential as an effective immuno-stimulating as well as an antidiabetic agent. For the purpose of enhancing EPS production, intensive strain improvement program was carried out by obtaining large amounts of protoplasts for the isolation of single colonies. Mass screening for high-yielding producers was possible through the formation of protoplasts because high concentration of protoplasts(2x106 protoplasts/ml) could be recovered by KOH-treatment of the producing mycelia with the cell wall disrupting enzymes. Especially, by obtaining variants with good fermentation properties such as low pigment production and high-shear resistance, 50-liter stirred-tank bioreactor cultures could be performed successfully in relatively high share environments. In the fermentation studies, production level of EPS was found to considerably depend on the medium compositions of both growth and production cultures. Especially, the ratio of the constituent carbon and nitrogen sources(C/N ratio) of the production medium was found to be one of the most important factors for the enhanced production of EPS.
