Mannopeptimycins: unique lipid II-targeted anti-infective agents

Bacterial natural products continue to be a rich source of potent antimicrobial agents. Our group recently described the application of new chemical and biological strategies in the discovery of the mannopeptimycins, a promising new class of glycopeptide antibiotics produced by Streptomyces hygroscopicus LL-AC98. These compounds block the lipid II dependent peptidoglycan maturation steps of Gram-positive bacteria by means of a mechanism distinct from other Lipid II-targeted agents. The unique chemical and biological features of the mannopeptimycins will be described, along with the results of studies to enhance the structural diversity and biological activity of these agents.
